Performance Validation
SYBR Green Method Validation (Gene Expression Detection)
HEK293 cells were enzymatically digested, washed with DMEM, counted, and distributed into six wells of a 96-well plate at cell densities of 15,000; 5,000; 1,667; 550; 183; and DMEM blank control. After centrifugation and supernatant removal, cells were lysed and subjected to RT-qPCR as per the kit instructions. Dark blue, light blue, dark green, light green, dark yellow, and light yellow bars represent the six sample groups. Panels A and C show amplification results for the UA kit and a domestic competitor’s product, respectively. Panels B and D display their corresponding melt curves. The UA kit demonstrates superior detection sensitivity and specificity compared to the competitor’s product.
TaqMan Probe Method Validation (Gene Expression Detection)
HEK293 cells were enzymatically digested, washed with DMEM, counted, and distributed into eight wells of a 96-well plate at cell densities of 15,000; 5,000; 1,667; 550; 183; 61; 20; and DMEM blank control. After centrifugation and supernatant removal, cells were lysed and subjected to RT-qPCR. The left-to-right order represents the eight sample groups. The negative control showed no amplification, while the 20-cell group exhibited faint signals at cycle 39. The remaining six groups showed clear amplification with excellent gradient linearity (Y-axis: Rn).
