PNGase F (Peptide-N-glycosidase F) is an amidohydrolase derived from Elizabethkingia miricola. It cleaves the amide bond of asparagine side chains (Asn-X-Ser/Thr, where X ≠ Pro), releasing intact N-glycans while converting Asn to aspartic acid (Asp). Unlike other glycosidases (e.g., Endo H), PNGase F can cleave all types of N-glycans (high-mannose, hybrid, and complex types) but cannot act on O-linked glycans or core α-1,3-fucosylated plant/insect N-glycans. This enzyme is widely used in glycoprotein research, biopharmaceutical quality control, and proteomics.
